Among the amino acids present, theanine is considered to play a pivotal role in the neuromodulatory effects of matcha. Furthermore, matcha retains the caffeine and various catechins, particularly epigallocatechin gallate, found in tea leaves. These active constituents collectively form the material basis for the regulation of central nervous system functions, exerting beneficial effects on cognitive function through synergistic interactions, such as theanine mitigating caffeine induced central nervous system over excitation and epigallocatechin gallate providing neuro antioxidant protection. Dietz and colleagues found through a series of cognitive tests that, compared to a placebo, participants who consumed matcha demonstrated significant improvements in attention and psychomotor reaction speed in response to external stimuli. Further research has confirmed that matcha exerts a protective effect on cognitive performance under specific conditions. In a randomized, placebo controlled trial, Baba and colleagues observed that healthy young adults who consumed matcha daily for two weeks maintained their attentional function, assessed via the Stroop test, significantly better when subjected to mild acute psychological stress, suggesting that matcha may alleviate stress induced cognitive decline. Regarding long term intervention effects, Uchida and colleagues conducted a twelve month randomized controlled trial involving elderly individuals with mild cognitive decline. The results showed that the matcha group performed significantly better than the placebo group in facial emotion recognition tasks, a measure of social cognitive ability, and also exhibited a trend toward improved sleep quality, confirming that long term matcha consumption may positively regulate social cognitive function and sleep status in the elderly. Notably, the ratio of active constituents in matcha is crucial to its neuromodulatory efficacy. Unno and colleagues indicated that the stress reducing effect is most pronounced when the ratio of the total molar amount of caffeine and epigallocatechin gallate to the total molar amount of theanine and arginine is less than two point zero. This finding offers a theoretical explanation for the variations in physiological effects observed among matcha products of different qualities or origins.

Research on cardiovascular and cerebrovascular protection indicates that matcha enhances angiogenic capacity, optimizes heart rate variability, and improves cardiovascular physiological function. Furthermore, the neuroprotective and cognitive enhancing effects of matcha have been confirmed in accelerated aging mouse models, healthy adults, and the elderly, with the underlying mechanisms involving tau protein regulation, improved synaptic plasticity, and enhanced attention and emotion recognition.

The various bioactive components in matcha play a positive role in improving cardiovascular and cerebrovascular health, regulating glucose and lipid metabolism, enhancing cognitive function, and maintaining gut homeostasis through multiple mechanisms, including antioxidant, anti inflammatory, metabolic regulatory, and neuroprotective actions, thereby providing a solid scientific basis for its development as a functional food. Although research into the health benefits of matcha has yielded significant results, certain limitations remain. First, clinical evidence is insufficient, as existing studies largely focus on animal models, lacking large sample, long term human intervention trials. Second, the mechanisms behind the synergistic effects of active components are unclear, given that the efficacy of matcha is often attributed to a single component, while the interaction networks among multiple components, such as the antagonistic balance between tea polyphenols, theanine, and caffeine, remain to be elucidated. Future research should prioritize overcoming the limitations of current animal models by conducting large sample, multicenter, long term human clinical trials to precisely define effective dosages, long term safety, and actual clinical benefits across different physiological states. Regarding component mechanisms, multi omics technologies should be integrated to deeply analyze the interactions among key components such as catechins, theanine, and caffeine, with a focus on clarifying how food matrices regulate the release kinetics and bioavailability of these active substances. Additionally, further research is needed on the interactions between characteristic gut metabolites of matcha and host metabolism, immunity, and neurological function. Investigating changes in the stability of functional components during processing and storage will provide scientific support for the standardized development and precise application of matcha based products.
